You can set one default language and allow users to select individual languges (among those you have made available in the languages folder). The default language is set in LocalSettings.php. After changing the language value, run the rebuildMessages.php maintenance script to update the system messages from the database.
http://www.mediawiki.org/wiki/Help:%24wgLanguageCode has a little information about the directive involved, and http://www.mediawiki.org/wiki/Help:Configuration_settings#Localization lists the functions related to Localization. Search seems to be broken on MediaWiki.org right now, so I can't actually locate the "walkthroughs" for localisation, but I know they're somewhere...
